I have  a baitcaster I want to spool the reel with powerpro 40lb braided line.

Should I put fluorocarbon on first to protect the spool or use electrical tape?

I wouldn't waste good fluorocarbon as backing. Use a cheap mono. The quality of backing doesn't affect your casting. You can use tape but you will use more braid then needed BUT you can also flip it next season and have brand new braid again. Either way will be fine.
